Photosystem II Repair Cycle in Faba Bean May Play a Role in Its Resistance to Botrytis fabae Infection

چکیده
Chocolate spot, which is caused by the necrotrophic fungus Botrytis fabae, a major foliar disease occurring worldwide and dramatically reducing crop yields in faba bean (Vicia faba). Although chemical control of this an option, it has serious economic environmental drawbacks that make resistant cultivars more sensible choice. The molecular mechanisms behind defense against B. fabae are poorly understood. In work, we studied leave proteome two genotypes respond differently to order expand available knowledge on such mechanisms. For purpose, used two-dimensional gel electrophoresis (2DE) combination with Matrix-Assisted Laser Desorption/Ionization (MALDI-TOF/TOF). Univariate statistical analysis gels revealed 194 differential protein spots, 102 were identified mass spectrometry. Most spots belonged proteins energy primary metabolism, degradation, redox or response stress functional groups. MS results validated assays protease activity gels. Overall, they suggest may different PSII repair cycle mechanism chloroplast. differences resistance be result metabolic imbalance susceptible genotype efficient chloroplast detoxification system at early stages infection.

fungal diseases, chocolate spot (botrytis fabae sard) is the most destructive diseases of faba bean and causes considerable dramatic damage losses on faba bean yield worldwide. the levels of resistance to chocolate spot were evaluated in 35 faba bean cultivars and one susceptible check (ilb365) and local check (barkat) at the gorgan agricultural research station during the 2010 and 2011 croppin...
متن کاملLarge-Scale Transcriptome Analysis in Faba Bean (Vicia faba L.) under Ascochyta fabae Infection
Faba bean is an important food crop worldwide. However, progress in faba bean genomics lags far behind that of model systems due to limited availability of genetic and genomic information. Using the Illumina platform the faba bean transcriptome from leaves of two lines (29H and Vf136) subjected to Ascochyta fabae infection have been characterized. De novo transcriptome assembly provided a total...
متن کاملComparative Analysis of Defense Responses in Chocolate Spot-Resistant and -Susceptible Faba Bean (Vicia faba) Cultivars Following Infection by the Necrotrophic Fungus Botrytis fabae
In this study, resistance responses were investigated during the interaction of Botrytis fabae with two faba bean cultivars expressing different levels of resistance against this pathogen, Nubaria (resistant) and Giza 40 (susceptible). Disease severity was assessed on leaves using a rating scale from 1 to 9. Accumulation levels of reactive oxygen species (ROS), lipid peroxidation and antioxidan...
متن کاملidentification of sources of resistance to the black bean aphid, aphis fabae scopoli, in faba bean, vicia faba l., accessions
in several parts of the world, important yield losses are caused to faba bean due to attack by the black bean aphid aphis fabae scopoli. although resistance could be a good alternative to chemical control, its genetic basis is still largely unknown; and its phenotypic expression is still poorly described, making crucial the permanent evaluation of genetic resources available in different countr...
متن کاملRevisiting the photosystem II repair cycle
The ability of photosystem (PS) II to catalyze the light-driven oxidation of water comes along with its vulnerability to oxidative damage, in particular of the D1 core subunit. Photodamaged PSII undergoes repair in a multi-step process involving (i) reversible phosphorylation of PSII core subunits; (ii) monomerization and lateral migration of the PSII core from grana to stroma thylakoids; (iii)...
